Male Anopheles mosquitoes are attracted to the flight tones produced by conspecific females as they beat their wings. This phonotactic attraction is essential for Anopheles mating to occur. Recently an Anopheles mutant line for the doublesex gene (dsxF) has been tested for future release as a control tool, which would suppress Anopheles mosquito reproduction in the wild. The effect of dsxF mutation on female flight tones, and thus courtship, is still unclear.
In this study, we assessed male and female dsxF mutant and control flight tones and used a phonotaxis assay to test acoustic preferences. We identified significant changes in dsxF mutant female flight tones compared to control females, though these remained distinct to males. Males showed stronger phonotactic responses to control female flight tones than to those of dsxF mutant females. No male mutants showed differences
from controls. In summary, although dsxF mutation does not directly affect males, the resulting changes in female flight tone can alter phonotactic behaviors related to courtship.
